2 Dimensional Gel Electrophoresis Rat RPE-J Cell Line

2D Gel from Proliferating RPE-J cells. RPE-J cells were grown at 32oC on tissue culture plastic and 2D gel analysis performed on protein extracts from whole cell lysates as described in Methods (~300 µg protein applied, 97,162 Vh IEF). Gels were stained with colloidal Coomassie Blue and numbered spots excised for protein identification. Circled spots are labeled by protein and differ in average staining intensity at least 2 fold relative to matched 2D gel spots; black circles denote greater intensity in proliferating RPE-J cells, red circles denote greater intensity in differentiating RPE-J cells. RPE-J cells were grown under differentiating conditions on Transwell filters at 40oC and 2D gel analysis performed on protein extracts from whole cell lysates as described in Methods (~300 µg applied, 97162 Vh IEF). Gels were stained with colloidal Coomassie Blue and numbered spots excised for protein identification. Circled spots are labeled by protein and differ in average staining intensity at least 2 fold relative to matched 2D gel spots; red circles denote greater intensity in differentiating RPE-J cells, black circles denote greater intensity in proliferating RPE-J cells.
